Capillary:

A crucial aspect of CE is the use of a narrow capillary tube, typically made of fused silica. This narrow inner diameter (typically ranging from 50 to 100 micrometers) provides several advantages, including reduced heat dissipation (Joule heating), increased electric field strength, and enhanced separation efficiency. The capillary is filled with an electrolyte solution that provides the necessary ions for the separation process.

Sample Introduction:

In CE, sample introduction is usually accomplished using a technique called hydrodynamic injection. A small volume of the sample is introduced into the capillary by applying a brief pressure difference, typically using a syringe pump. This pressure-driven injection ensures sample introduction without significant band broadening, preserving the separation efficiency.

Detection:

Detection in CE is typically achieved by monitoring the changes in absorbance of the separated ions as they pass through a detection window in the capillary. The detection window is usually located near the end of the capillary, allowing for the detection of analytes as they elute from the capillary. Various detection methods can be employed, including UV-Vis absorbance, fluorescence, and mass spectrometry.

Data Analysis:

The output of a CE analysis is an electropherogram, which is a plot of the detector signal intensity versus the migration time of the analytes. Each analyte produces a peak in the electropherogram, and the peak area or height is used for quantitative analysis. The migration time of each analyte provides qualitative information for analyte identification.

Applications of Capillary Electrophoresis:

Biological Molecules:

CE is widely used for the separation and analysis of biomolecules, such as proteins, peptides, and nucleic acids. It is particularly useful for analyzing complex mixtures due to its high separation efficiency and sensitivity.

Pharmaceutical Analysis:

CE plays a significant role in the pharmaceutical industry for quality control, purity assessment, and drug formulation studies. It is used to analyze active pharmaceutical ingredients, impurities, and excipients.

Environmental Monitoring:

CE finds application in environmental monitoring for the analysis of pollutants, heavy metals, and other contaminants in water, soil, and air samples.

Food and Beverage Analysis:

CE is employed to analyze food additives, sweeteners, preservatives, and contaminants in food and beverage samples.

Forensic Analysis:

CE is used in forensic science to analyze DNA samples and detect the presence of drugs and toxins in biological fluids.
